By thus inflating the likelihood of observing a false-positive association, multiple testing may compromise a study’s validity. Furthermore, even where this is not the case, modeling multiple congeners requires multiple statistical tests, presenting many opportunities to make erroneous inferences by chance. However, oversaturating the model with dozens of congeners may result in high uncertainty in model estimates and possible non-convergence or model failure. Disentangling their individual associations entails modeling all compounds together to control for possible confounding. Firstly, blood levels of these congeners are highly correlated and may confound associations of single compounds with a given health outcome ( Covaci et al., 2002 Longnecker et al., 2000). The large number of PCDDs, PCDFs and PCBs presents challenges in studying their health effects. In particular, they may alter the timing of puberty in children and diminish fertility later in life ( Attfield et al., 2019 Greenspan and Lee, 2018 Mínguez-Alarcón et al., 2017 Sergeyev et al., 2017). PCDDs, PCDFs and PCBs are known to exert endocrine effects in rodents and may affect human reproductive function ( Bergman et al., 2012 Diamanti-Kandarakis et al., 2009 Gray et al., 2001 Meeker and Hauser, 2010 Rogan and Ragan, 2003). The most toxic dioxin, 2,3,7,8-Tetrachlorodibenzodioxin (TCDD), has a half-life of approximately 7 years in humans, while non-dioxin-like PCBs have half-lives ranging from approximately one month for PCB 77 to 22 years for PCB 189 ( Milbrath et al., 2009). Once in the human body, they persist for months to years, metabolized slowly at rates that depend on age and on the number and position of the chlorine atoms in their molecules ( Grandjean et al., 2008 Grimm et al., 2015 Milbrath et al., 2009 Quinete et al., 2017). These lipophilic chemicals are deposited into the environment, where they accumulate in the food chain and in human adipose tissue ( Béchaux et al., 2014 Hoogenboom et al., 2015).
